Is HostZERO Microbial DNA kit compatible with DNA/RNA Shield?

Samples stored in DNA/RNA Shield are not compatible with our HostZERO Microbial DNA Kit. This kit is optimized for fresh samples and not for samples that are immersed in lytic solutions (Shield is lytic to an extent, lysing easier-to-lyse samples).

How long should I bead beat my sample for?

Please refer to the “Optimized Lysis Protocols” document for our validated instruments and protocols. Additional optimization may be needed

If I have my samples stored in DNA/RNA Shield, do I need to add ZymoBIOMICS Lysis Solution?

Answer 3

No, if your samples are stored in Shield you should not add the ZymoBIOMICS Lysis Solution. You can homogenize directly in Shield

Is the HRC filter step necessary?

Answer 4

The Zymo-Spin III-HRC Filter removes PCR inhibitors from your sample. Certain sample types like soil, feces, and plants contain high amounts of inhibitors, so the HRC step is strongly recommended. Other sample types like cultured bacteria or yeast do not strictly need the HRC filter step.
