How do I reconstitute Proteinase K?

A

For 5mg of proteinase K, please reconstitute it in 260µL of the storage buffer. For smaller or larger formats, you can scale up or down the volume of the storage buffer proportionally if you want to keep the concentration the same. The concentration should be 20 mg/mL.

Does bead beating cause shearing?

A

Yes. On average, post bead beating, genomic DNA is between 15-20 kb depending on the initial quality of the sample, making it amenable to Next-Generation Sequencing platforms requiring high molecular weight DNA. For optimal DNA integrity, collect samples in DNA/RNA Shield™.

Are the Zymo-Spin I columns interchangeable with the Zymo-Spin ___ columns?

A

Yes, most of our columns are interchangeable with the exception of the ZymoPURE and Select-a-Size kits. The binding capacity and elution volumes are different, but the columns can be used in any of our Quick-DNA/RNA, Direct-zol, and clean-up kits

A

The IICR is an updated version of the IIC and contains a retention ring for improved matrix stability. Otherwise, the columns are the same.

What is the difference between a capped and uncapped column?

A

The only difference is the cap. Everything else between the columns (max capacity, column matrix, etc.) is the same. Unsure which to use? It mainly comes down to preference. Some people like the capped columns for easy labeling or added security while others like the uncapped columns for faster sample processing.

My kit is expired, is there anything I can still use?

A

If your kit is expired, we would recommend purchasing a new kit for optimal results. However, the wash buffers should be relatively stable if capped tightly. Columns have to be replaced after the expiration date as binding efficiency decreases with age.*
