media and stains Flashcards
what is SABs primary isolation for
fungus
SABs pH
6; makes it inhibitory for some bacteria
basic nutrient media, no antibiotics
SABs + blood is good for
sporulation of some of molds
BHI+ (brain heart infusion) antibiotics added
cycloheximide and chloramphenicol; keep down bacteria
BHI referred to as ??
mycosel plates or BHI with CC plates
corn meal/ potato dextrose agar
grows mold and yeast and used for SPORULATION
-enhances pigment of any fungus
birdseed agar
- Detect presence of Cryptococcus neoformans
○ Produce brown pigment
chromager
- Really used for ID yeast
- Cornmeal base to which indicators have been added allowing different yeast types to produce different colors
KOH Prep
- Determine presence of a fungal element on hair, skin, and nails
- Placed in KOH solution and sit for 15-30 min in order to dissolve keratin
○ Allow to see fungal element - ONLY tells you if something is present
- Placed in KOH solution and sit for 15-30 min in order to dissolve keratin
calcofluor white stain
- Fluorescent stain that can be used in place of KOH; more sensitive
- Fungal elements in hair, skin, and nails
- Stain binds to chitin on fungal element allowing to fluoresce
india ink prep
- Detect large capsule Cryptococcus neoforms
○ Placed on slide and India Ink dropped onto specimen and viewed
gomori silver nitrate stain
- Detect presence of Pneumocystis jiroveci
- # 1 cause of death in HIV/AIDS patients
- Common specimen = lung tissue
Periodic-Acid-Schiff Stain (PAS
- Detect presence of fungal elements in vivo (tissue specimen, biopsies, etc.)
- Not used in clinical lab
- Fungal elements will stain pink or purplish in specimen
Lactophenol Cotton Blue (LPCB)
- Used to help visualize fungus under microscope
- Helps visualize components better, see different structures
Used in vitro
- Helps visualize components better, see different structures
gold standard for looking at sporulation structures
slide culture technique
slide culture technique
enhance sporulation of fungus/ molds
square of PDA or CMA
-piece of growing fungus placed on 4 corners with teasing needle and place coverslip on top
-incubate, and after a couple weeks place coverslip into LCPB to view structures
scotch tape prep
- Take clear piece of scotch tape, place into a growing fungi and then place into LPCB
teasing the fungus
- Tease off growing colony with needle
Need to sterilize in between uses
yeast API
- Yeast Identified by sugars utilized
- If uses sugar turns turbid
- ONLY yeast, no mold
Sugar assimilation (means uses the sugar)
