Lecture 7: Cytoskeleton IV Flashcards
Name the three types of mitotic MT
-astral MT
-kinetochore MT
-Polar MT
Astral MT function
connect the spindle to membrane to ensure the spindle position
Kinetochore MT function
attach to kinetochore to pull chromosome apart
Polar MT function
use kinesin motor to push the two spindle poles apart
Describe FRAP
Fluorescence Recovery After Photobleaching
-helps determine how fast the GFP signal recovers at the site of photobleaching
XMAP215
-stabilizes (+)
-promotes MT growth
-regulated by phosphorylation
Name the drug used as an anti cancer drug
Taxol
What does taxol do?
It binds to microtubules and inhibits microtubule depolymerization and dynamics- inhibits mitosis so the cancer cells will be killed due to mitotic effect
What is tightly controlled during cell cycle?
centrosome number
Why is it important to tightly control centrosome number?
Too many centrosomes:
-chromosome segregation defects
-genetic instability
-tumorigenesis
What do centrosome abnormalities suggest
cancer
Intermediate filaments support
nuclear structure (lamins)
Lamin related disease
Hutchinson Gilford Progeria Syndrome (HGSP)
List three things about IF
-dynamic
-assembly occurs spontaneously
-energy is required for disassembly
Which phase does duplication happen?
S phase
