Gel electrophoresis
Method to seperate DNA molecules based on their size using an electric field to move molecules through a gel matrix
The gel used:
Agarose polymer: forms complex matric of pores
Negative charge is applied to
the top of the gel where the DNA is added to the wells
Positive charge is applied to the
bottom of the gel
DNA charge is… because…
negative because of the phosphates
Small DNA molecules will travel
quickly through the gel matrix
Large DNA molecules will travel
slowly through the gel matrix (Stuck in pores)
Molecular ladder
Contains DNA pieces of know sizes to help determine the sizes of your DNA samples
DNA travels at a rate…. this is why the spacing…
- that is inversely proportional to the log of its size
- the space between 2000->1000 isnt the same as 200->100 because DNA travels at a rate inversely propotional to the log of its size
Plasmids can be found in 3 different conformations
- supercoiled
- circular relaxec (nicked)
- Linear
