lecture 20 redone Flashcards
ON mechanism cAMP production
cAMP production is via the enzyme
adenylyl cyclase
there are 2 different types of adenylyl cyclase
- in the plasma membrane
- soluble enzyme exists in the cytosol
what domains to type 1 have
2 TMDs and 2 catalytic domains (C1 and C2)
in the off state the catalytic domains are
separated
when activated the catalytic domains
are together
when activated the ATP can be converted into
cAMP
type 2 is activated by
increased HCO and Ca
forskolin from plant alkaloid
actiavtes the enzyme
OFF mechanism cAMP breakdown
phosphodiesterase (PDEs)
cilastazol is a
PDE3 inhibitor for peripheral vascular disease (cAMP vasodilation to improve blood flow)
CFTR agonist example
adenosine
AKAPS bind to
PKA
2 types of PKA
type II can bind to AKAPs it has a docking domain which it can bind to, AKAPs bring the sibstrate close to the catalytic subunits in type II
HT31 peptide disrupts
PKA-AKAP interaction
ezrin (AKAP) targets PKA to CFTR this requires a
scaffold protein (NHERF1)
