Explain the ‘shotgun approach’
A
- Cut DNA from many copies of an entire chromosome into overlapping fragments
- clone the fragments in plasmid or phage vectors
- sequence each fragment
- order the sequences into one overall sequence

How do you find genes by their function?
A
isolate the mRNA’s that genes make and sequence them to reveal their intron/exon structure

What are open reading frames (ORF)
A
sequence of codons that do not contain a stop codon

what genes can be hard to find ?
A
- small genes
- overlapping genes
- intron/exon splice sites

How do you find genes by homology ?
A
- take human and mouse genes
- look for tracts of sequence that share homology
- Sequencies in common imply: they are important and have a function

Why are homeotic genes key ?
A
they are good for determining positional identity along the length of an organism
