How to prepare a wet mount?

Answer 1

1. Dry and clean slide
2. Circle on the opposite side
3. Label slide (e.g. SA, EC)
4. Add H2O or saline
5. Add loopful of bacteria using inoculating loop (aseptic transfer)
6. Heat loop, let it cool
7. Mix with water
8. Place cover slip

Glass slide -> inoculating loop with broth or water then slant -> cover slip (sometimes heat fix) -> microscope
