Q
Why does DNA precipitate in NaCl and ethanol
A
NaCl neutralizes DNA reducing its reaction with water molecules.
Ethanol is less polar than water therefore DNA precipitates.
1
Q
EDTA
A
Chelates metal ions which are cofactors for DNases. Also in anticoagulant for blood sample.
2
Q
High 260/280 ratio
A
High pH of sample
High levels of RNA
3
Q
Low 260/230 ratio
A
EDTA
Carbohydrates
Some proteins.
4
Q
Sucrose
A
Increase solution osmolarity - cells burst
5
Q
Triton X-100
A
Detergent that solubilises membrane proteins and lipids.
Destroys mitochondria and organelles.
6
Q
What does SDS do?
A
A negatively charged detergent that binds proteins.
Dissociates multimeric proteins and causes polypeptide chains to denature .
