LAB - Kinetic Spectrophotometry... Flashcards
1
Q
this helps us measure enzyme activity in order to quantify them
A
kinetic assay = measures rate of rxn
2
Q
gamma-Glutanyktransferase
A
catalyzes transfer of glutamyl group from a glutamyl peptide to sn AA of another peptide
- several organs but increases in serum are usually from hepatobiliary system
- used in diagnosis for liver diseases such as alcoholic cirrhosis and secondary liver tumors
- this may be used to confirm if an increase in ALP originate in the liver
3
Q
endproduct of the reaction y-GT catalyzes which absorbs strongly at 405 nm
A
5-amino-2-nitrobenzoate
4
Q
KINETIC assays
A
- enzyme incubated with substrate
- substrate, product, co-factor measured continuously or several points as a function of time
- allow for demonstration of linearity of a rxn
- measured over a short period of tie = little danger of enzyme inactivation
- warning provided if substrate conctn falls below optimum (NOT in fixed time or end-pt rxn)
