Q

List 3 ways to care (safety) for the microscope?

A
  1. carry by holding the base and arm (microscopes are top heavy)
  2. only use lens tissue and cleaner
  3. Dont let the cord drop to the floor
Additional - reset scanning objective in place (5x), lower stage, wrap cord

16
Q

what is the function of the ‘Iris Diaphragm lever’?

A

increase and decrease light to the specimen

17
Q

what is the function of the ‘lamp’?

A

light source

18
Q

what is the function of the “on and off switch”?

A

turns lamp on and off

19
Q

what is the function of the ‘brightness control knob’?

A

increase and decrease light from the lamp

20
Q

what is the function of the ‘Revolving Nose Piece’?

A

support objective lenses, allow changing of lenses

21
Q

what are the objective lenses?

A

4 lenses, used to magnify objects

22
Q

1.what is the function of the ‘Eyepiece’ (ocular)?

2.what is the power of the ocular lens?

A

1.contains the ocular lens
(closes to the eye-one eyepiece may have a pointer)

2.has a power of 10x - shows the item 10 times bigger

23
Q

what are 3 rules to follow when using the microscope?

A
  1. never use the immersion objective without immersion oil
  2. always begin focusing with the scanning objetive and stage all the way down
3.never adjust the coarse adjustment knob while using low or higher objective lens
24
Q

Microscope Basics - FACTS

A

the image is turned upside down and reversed due to the objective lens, a mirror and the ocular lens.

26
Q

What is “Total Magnification”?

and how does it works?

A

the power of the objective lens multiplied by the power of the ocular lens

Total Mag = Ocular power X Objective power

Eg: Using high power: 10 X 40 = 400x
(10 ocular lens ‘multiplied by’ 40 high power lens = 400 times total magification)

27
Q

what is ‘field of vision’?

A

Amount of specimen and surrounding area that can be seen.

(it changes with each objective)

28
Q

TRUE OR FALSE?
1.The field of vision DECREASES with inreasing magnification?
2. Amount of details INCREASES with increasing magnification?

A

1.TRUE
2. TRUE

29
Q

what is ‘Parfocal lenses’?

A

Allow objective lenses to stay in focus

NOTE - if switched from scanning-to-low power-to-high power the object stays in focus
5x»10x»40x (fine adjustment should only be needed)

30
Q

TRUE OR FALSE?
The coarse adjustment knob is not needed when using objectives greater than the scanning objective

A

TRUE

31
Q

whis is “working distance’?

A

distance between the stage/specimen and the objective lens

32
Q

what is ‘Depth of view’?

A

ability to distinguish closer and deeper structures

33
Q

what is a ‘Binicular’?

A

instrument that allows the use of both eyes

34
Q

STEPS IN FOCUSING USING THE SCANNING OBJECTIVE

A
  1. stage should be all the way down
  2. scanning objetive should be in place
  3. Place the slide on stage
  4. Center and adjust light
  5. Raise stage all the way up
  6. Focus downward using the coarse adjustment knob
  7. Center and adjust light
  8. Fine focus using the fine adjustent knob
  9. center and adjust light (if necessary)
35
Q

STEPS TO REMOVE SLIDE AND FINISH

A
  1. Rotate scanning objective in place
  2. Lower stage using coarse adjustment knob
  3. Remove slide
  4. Turn off lamp
  5. Unplug cord and wrap around the microscope
  6. Put back in place on shelve 3x3x3x4
