The microorganism (microbe) has to be in large amounts in the cases of the disease, but it cannot exist in abundance in healthy organisms.

The microbe has to be singled out and put in a controlled lab environment for it to grow. By isolating the microbe and putting it in a lab, epidemiologists can study the cause and effect relationship between the microbe and the disease more in depth.

The isolated microbe in the lab should cause the disease once put into a healthy organism.

The microbe has to be re-isolated after its “installment” into the healthy organism. Then, study the disease in the experimental host and see if it’s similar or identical to the disease you’re looking for.
