Jordans Notes 3 Flashcards
What are the two steps to cause lysis?
- Tissue maceration (mechanical breakdown)
- Heating in buffer (increases cell breakdown) -> detergent, proteinase K, chaotropic salts
What are the beads that DNA wrap around?
Histones
If you precipitate a solution, what are you doing?
Causing a solid substance to separate from a solution
A group with an ancestor but not all descendants?
Paraphyletic
- Reptilia without including birds
- - lizards without snakes
What is the liquid left over from precipitation/centrifugation?
Supernatant
Which DNA extraction has no separate lysis and purification step?
Chelex extraction
What temperature is ok for short term DNA storage?
4 degrees C refrigeration
How many phosphate groups does a nucleotide have?
1
If you don’t know what species may be present in a sample, what kind of PCR primers should be used?
Universal primers
Left over unused primers in PCR product
Primer dimers
not a huge issue
