Convenient
Easily subjected to controlled changes
What are constituents in organ bath?
Physiological buffer (water based)
Physiol salt solution: Ringers, Tyrodes,Krebs Haseleit
Aeration: 95% oxygen and 5% carbon dioxide
How should tissue be collected?
Fast dissection
Minimal handling
Dissected/transported on ice
What tissues deteriorate fast?
Liver
What tissues are stored in chilled buffet at 4oC in the fridge?
Guinea pig ileum (aerate)
Smooth muscle strips & rings
Papillary muscle
What are limitations of isolated tissue preps?
Damage in isolating
No exact match to internal conditions of body
Delivery of oxygen & nutrients by diffusion (hypoxic core)
