Immunomodulatory Drugs Flashcards
What are cytotoxic agents.
Kill rapidly dividing cells.
Acronym for cytotoxic agents.
MAC:
Methotrexate
Azothioprine
Cyclophosphamide
Methotrexate MOA
Folic acid antagonist (inhibiting DNA and RNA synthesis).
Stomatitis side effect.
Azothioprine MOA
Inhibits purine and DNA synthesis.
Stomatitis side effect.
Cyclophosphamide MOA
Broken down to cytotoxic mustard and acrolein.
Cytotoxic mustard cross-links DNA, leading to cell death.
Acrolein causes hemorrhagic cystitis in the lower urinary tract.
Types of T cell inhibitors.
Cyclosporine
Tacrolimus
Cyclosporine MOA
Cyclosporine binds to cyclophilin, which inhibits calcineurin to suppress the immune system post-transplant.
Tacrolimus MOA
inhibits T cells without bone marrow suppression.
Prevents rejection of solid organ transplant, tropical tx of eczema.
Low doses are used for replacement therapy, while high doses are used for immunosuppression.
Glucocorticoids.
Glucocorticoid used for replacement therapy.
Hydrocortisone
Glucocorticoid used for immunosuppression.
Prednisone and Dexamethasone
