HTLV1 and Prions Flashcards
HTLV1:
delta retrovirus, oncogenic
HTLV1:
Structure – 2 ssRNA, icosahedral, enveloped
Epidemiology – Caribbean, Japan, West Africa
HTLV1:
Replication – reverse transcriptase → genome integration → replication → virus stays in the cells, PIC transferred to adjacent cell
HTLV1: Adult T cell leukemia
– floral nuclei T cells, skin lesions – acute, chronic, smoldering, lymphoma
HTLV1: Tropical spastic paraparesis/myelopathy (TSP/HAM)
- ↑ infected T cells, cytokines and CD8 → Tax → demyelination
Tax protein
– gene expression, inhibition of apoptosis, activates growth factors, inhibits DNA repair
CD8 T cell target → ↓ proliferation, Tax protein loss, HBZ expression → floral nuclei, disease
HBZ
– stimulates T cells production (E2F1), down regulation of Tax
Prions:
case transmissible spongiform encephalopathies
Prions: Histo
– vacuolation of neurons, amyloid aggregates, atrophy w/ inflammation
Bovine spongiform encephalopathy
– aka Mad Cow, variant Creutzfeldt-Jacob disease
Scrapie protein
– β sheet dominant, insoluble, resistant to proteinase K degradation, recruits other normal proteins
