Fear Conditionning Experiments
Stimulate in the medial geniculate and record in lateral amygdala
Electrical stim - HFS - electrical epsp = increase response for same stim = LTP
Auditory epsp - HFS - ausi epsp = increase response = LTP
Parralel mechanisms
Food pellet training
Rat trained to retrieve food from right hand. Removed brain, stimulated and recorded
Motor learning is controlateral
Control; response is the same for trained and untrained hemisphere
Trained rats= response is bigger for trained (left) hemisphere
NMDAR antagonist AP-5 and LTP
When the slice is treated zith AP5, LTP cannot be induced.
The response for a bigger stimuation is bigger, but then it goes back to the same level when weak stimuation
AP5 and Morris WM
Gave AP5 or vehicule and then trained them to find hidden platform
Rat with AP5 didn’t learn, less time searching in target quadrant when probe
Trained animals and then gave them AP5
Rats learned how to find the platform
- NMDAR - LTP - Learning = related
- NMDAR - induction of LTP only
Doogie mice
Add 1 gene to increase NMDAR
- Slice exp show enhaced LTP
- Behav exp show improvment, not huge but present
- after may training, the result is the same for all animals
Dissociation of LTP and learning
ACPD is a mGlutR agonist that blocks LTP
When animal are treated with ACPD it _doesn’t impair learning _
