How do you carry out an ABO and RhD type in tubes
Need patients rbcs and plasma
Anti A
Anti B
Anti AB
Anti D1
Anti D2
Rh Control (RC)
A1 cells + plasma
B cells + plasma
Incubate at room temp for 5-10 mins
Centrifuge at R2
How do you carry out an ABO and RhD type in gels

Get a normal gel containing anti A, B, D and control
Add 10ul patients rbcs to each well
Add 50ul reagent rbcs to reverse groups (A1 and B) then add 50ul patient plasma
Centrifuge for 10 minutes
How do you carry out a Rh phenotype in tubes
Patient rbcs, reagent rbc controls and anti sera
Anti D
Anti C
Anti c
Anti E
Anti e
Anti K
+ control reagent Biorad cell
- control reagent Biorad cell + anti-sera
Incubate for 5 mins
Centrifuge at P5
Rh phenotyp in gels
Find ID card with anti C,c, E, e, K and control present
Add 10 ul (3-5%) patient rbcs to each well
Centrifuge for 10 mins
3 panel antigen test

You need x2 LISS/Coombs Gels (Green lines on gel) -> one for your patient and one for your controls
You need 3 BioRad reagent cells (I, II, III)
Controls = 25ul weak anti-D or 25ul Inert AB to each reagent cell (50ul) on control gel i.e. no patient
Needs to be incubated for 15-20 mins at 37 degrees
Antibody identification
You need x2 LISS/COOmbs gel cards (green)
You need x2 Enzyme Panel - NaCl Gels (red)
You need your BioRad Antibody ID panel - untreated (white cap)
You need you BioRad antibody ID panel - papanised (red cap)
Need to add 50ul of each rbc and 25ul patient plasma to each
+ Control needed for untreated and treated panel -> 25ul weak anti-D with 50ul RhD positive cell (pick from panel)
Need to incubate at 37 degrees for 15 minute
