Enzyme Kinetics Flashcards
Michaelis menten kinetics: types of reactions
1st order reaction
pseudo 1st order reaction
zero order reaction
1st order reaction
pseudo 1st order reaction
zero order reaction
1st order reaction: rate = [S], the concentration of a single substrate is directly proportional to the rate of the rxn. linear relationship between [s] and product formation
pseudo 1st order reaction: only one substrate effects the rxn rate. have multiple substrates but one is already fully saturated the enzyme
zero order reaction: the rate stops increasing even though the [S] is increasing because enzymes are saturated
what are the 2 things being measured in michealis-menten equation
vmax= maximum rate
km= enzyme affinity
Michaelis-menten equation
Vo= Vmax[s] / ([s]+Km)
Vo= initial velocity, initial rate of rxn. When [s] is high and [p] is low there’s no reverse rxn
Vmax= maximum rate/ velocity
Km= enzyme affinity. indicates how well the enzyme binds a given molecule (inverse relationship)
small km= high affinity
large km= low affinity
line weaver-burk
reciprocal of Michaelis menten, straight line graph
what are the x intercept and y intercept in line weaver-burk plot
x intercept = -1/Km (NEGATIVE)
y intercept= 1/Vmax
higher Vmax is lower y intercept
higher Km, less affinity = largest x value
lowest Km, highest affinity= most negative X value
