ELM6: Drug target bonds Flashcards
What is the structure of a proteinogenic amino acid?
Amino acid attached to an alpha carbon
Alpha carbon always chiral
All L form
What is an R group?
Varies for each amino acids
Can be 1 of 21 different side chains
What sort of reaction forms a peptide bond?
Condensation
Where is the peptide bond between amino acids?
Between carboxylic acid group on first amino acid and amino group of the second
What is the N terminus?
The free amino acid
What is the C terminus?
Free COOH group
What is the binding domain?
Part of a protein a drug interacts with
What is the importance of protein folding and binding domains?
Domain might have several interaction points
When in a chain they might be far away and only get closer when the protein folds
What is a pharmacophore?
Minimum set of structural features a molecule must have to fit into a targets binding domain
What is the use in working out the pharmacophore of a target?
Can search databases for molecules that fit and use them in drug development
What are cation aromatic interactions?
Cations interact with electron cloud of an aromatic amino acid side chain
Binding of acetylcholine to nicotinic acetylcholine receptors
What are covalent bonds?
Sharing electrons in the valence shell
Strong
What are ionic bonds?
Charge charge interactions between anions and cations
What are hydrogen bonds?
Two electronegative atoms share a hydrogen
Atoms are in a straight line
What is the H bond donor?
Atom hydrogen belongs to
