Effect of ethanol conc on membrane permeability Flashcards
1
Q
What is the permeability of a membrane measured by?
A
Beetroot cells, which contain a purple pigment called betalain
2
Q
What are cell surface membranes made up of?
A
Phospholipid bilayer, which makes them partially permeable
3
Q
How can permeability be changed?
A
By solvents and temp
4
Q
What happens when the membrane has a higher permeability?
A
More pigment leaks out of cells
5
Q
What is the relationship between concentration and absorbance?
A
As the concentration increases, absorbance increases since cells gain a higher permeability
6
Q
Describe 1st part of method: Prep and leaving for 15 mins
A
- Get 6 test tubes and put the different concs in them - 0%, 20%, 40%, 60%, 80%, 100%
- Trim beetroot cylinders to 5mm + rinse with water then pat dry
- Add 1 cyclinder to each tube + leave for 15 mins
7
Q
Describe 2nd part of method: Measuring absorbance
A
- Swirl tubes + use forceps to remove cylinders
- Pour remaining liquid into 6 cuvettes, fill another with distilled water
- Distilled water will act as blank for colorimeter
- Use colorimeter with green filter (520nm) to measure absorbance (AU) for each conc
- Record results in table + plot graph
