Synthesis
• Synthesized in mesencephalon (midbrain) in the substantia nigra and ventral tegmental area
Nigrostriatal Pathway
o Substantial Nigra-> striatum
Movement
Mesolimbic pathway
o Ventral tegmental area-> limbic system (motivational behavior and reward)
Learning
Mesocortical Pathway
o Ventral tegmental area-> prefrontal cortex (higher cognitive functions)
Attention and Learning
Parkinson’s Disease:
- L-Dopa used to treat Parkinson’s because it can cross blood brain barrier
- Limited efficacy because of side effects, negative feedback
Subfamilies:
- D1 Like- D1, D5
* D2 like- D2,3,4
Amphetamines
o Agonist, blocks reuptake of dopamine
o Not specific, also enhances norepinephrine
o Helps with disorders of excessive sleep
Antischizophrenic (Antipsychotic drugs)
o Antagonists, blocks D2 receptors
o i.e. Haloperidol
o Can cause motor side effects (one neurotransmitter has many functions)
