Dehydration Flashcards
The best dehydrating agent (fast acting; mixes with water and organic solvents; penetrate tissue easily)
Ethanol
Used if good-grade absolute ethanol is not easily available
Isopropanol
Toxic; for blood and tissue films and for smear preps
Methanol
For plant and animal microtechniques
Butanol
Cheap, rapid dehydrating agent; for utgent biopsies; volatile: flammable
Acetone
Excellent dehydrating and clearing agent; miscible to water, melted paraffin, alcohol, and xylene; expensive; toxic
Dioxane/Diethylene dioxide
Dehydrates rapidly; not harmful to tissue; toxic by inhalation, skin contact and ingestion
Cellosolve/Ethylene glycol monoethyl ether
Tissues can be transferred directly after fixing and washing; dehydrate sections and smears following certain stains
Triethyl phosphate
Dehydrates and clears tissues; miscible to water and paraffin; toxic; eye and skin irritant; cause conjunctival irritation (6 months exposure)
Tetrahydrofuran
3 additives that are added to dehydrating agents in duly hard tissues
1) 4% phenol
2) Molliflex
3) Glycerol alcohol mixture
