Clearing Flashcards
Ideal amount
10x
Used when the tossue is cleared directly from water (frozen section)
Glycerin/Gum syrup
Excellent and true clearing agent; most rapid; can be used with celloidin sections
Xylene
Substitute to xylene or benzene; nit carcinogenic; slower than benzene; expensive
Toluene
Rapid actung; for urgent biopsies and routine pruposes; higly flammable; carcinogenic; damage bone marrow causing aplastic anemia
Benzene
Best of the traditional clearing agents for routine use; slower than xylene but less brittleness; widest latitude; for tough and large specimens; best for delicate and higly cellular specimens; does not make tissues transparent; hepatotoxic
Chloroform
Clears both paraffin and celloidin sections; for CNS and cytological studies of smooth muscle and skin; very expensive (2 changes only); quality is not uniform; extremely slow; milky
Cedarwood oil
For clearing embryos, insects, and very delicate specimens
Aniline oil
Minimum shrinkage, removes aniline dyes; dissolves celloidin; tissues become brittle; tendency to be adukterated; expensive
Clove oil
Similar to chloroform but cheaper; toxic
Carbon tetrachloride/CCl4
Dehydrates and clears at the same time
Tetrahydrofuran
Slow acting; used for double embedding trchniques
Methyl benzoate; Methyl salicylate
