Staining constituents of cells to help determine lineage; used when cells are so immature that you cannot identify them from morphology alone
Cytochemistry
Enzyme found in primary granules of neutrophils
- Positive in myeloid cells
- Negative in lymphoid cells
Myeloperoxidase stain (MPO)
Stains lipids and is slightly less sensitive than MPO
- Positive in primary and secondary granules in neutrophils and monocytes
- Negative in lymphoid cells
Sudan Black Stain (SBB)
Primarily used to identify early granulocytic cells where red indicates positive
- SPECIFIC ESTERASE
Naphthol-AS-D-Chloroacetate Esterase (NASDA)
Primarily used to identify monocytic cells
- NONSPECIFIC ESTERASE
Alpha Naphthyl-Butyrate/Acetate Esterase (ANBE/ANAE)
