Good sample collection technique?
Aseptic
Refrigerate samples immediately
Wait 1-2 wk post antibiotics for good results
Gram Positive results appear?
Blue/Purple - easy to stain and hard to decolorize
Gram Negative results appear?
Red - picks up counter stain after decolorizing alcohol
Gram Variable results appear?
Red and blue mixed together. Can be due to over decolorization or old gram positive bacteria that dont take on stain
Nutrient Media is used for?
Basic growth of many pathogens.
Selective media is used for?
Facilitate the isolation of particular bacteria
Bacteria turn brilliant agar green with the presence of?
Salmonella
MacConkey agar does what?
Suppresses gram positive, thusly promotes gram negative
Mannitol salt agar does what?
Not used often but highly selective for Staphylococcus

Blood agar

Beta-hemolytic indicates what?
Can utilize all RBC for energy causes clear zone
Alpha-hemolytic indicates what?
Utilizes partial breakdown of RBC and greenish hazy appearance
Gamma-hemolytic indicates what?
Bacteria used no RBC and just grows on top
MacConkey indicates what?
pH indicator - turns pink as lactose is broken down
indicates E. Coli and Klebsiella
Bile salts - selective for enterobacter
