Chromatography Flashcards
What can chromatography be used for?
To separate and identify components of a mixture
What are three types of chromatography? (mentioned on spec)
Thin-layer chromatography (TLC)
Paper chromatography (PC)
Gas liquid chromatography (GLC)
How do you calculate Rf values? (chromatography)
Rf = distance moved by component / distance moved by solvent
What does separation depend on? (chromatography)
The balance between solubility in the moving phase and retention by the stationary phase
How does thin layer chromatography work?
A plate is coated with a powdered solid and a solvent moves up the plate
How does gas liquid chromatography work?
A column is packed with a solid or with a solid coated by a liquid, and a gas is passed through the column under pressure at high temperature
What is gas liquid chromatography commonly used for?
To separate mixtures of volatile liquids
What is the retention time of a substance? (chromatography)
The time taken for a particular compound to travel through the column to the detector
How does column chromatography work?
A column is packed with a solid and a solvent moves down the column
