Binding of Topo-II-DNA complex which leads to double-strand breaks
Free radical production via semiquinone radical intermediate, significantly increase in the presence of Fe
Doxorubicin Thera use
AML, ALL, HD, KS, Breast, Neuroblastoma
Doxorubicin side effects
BMS (dose limiting), Extravasation (tissue necrosis), Stomatitis, Alopecia, 2o AML, Cardiotoxicity: arrhythmia, CHF, tx Dexrazoxane, cardioprotective Fe chelator

MDR, increases glutathione peroxidase, decreases TOPO-II activity
Flutamide MOA
Androgen receptor antagonist
Flutamide thera use
Prostate (usually combined w/ leuprolide)
Flutamide side effects
Gynecomastia, Hot flashes, ↓ libido, Hepatotoxic
Aldesleukin MOA
Human recombinant interleukin-2, increases immune system, e.g., ↑ TNF, IL-1, interferon–γ, which inhibits tumor growth
Aldesleukin thera uses
RCC, Melanoma
Aldesleukin side effects
Capillary leak syndrome (hypotension, decrease organ perfusion, edema), Infections, BMS (RBC, Platelets), Mental status changes (irritability, confusion, depression), Thyroid fxn impairment, Arrhythmias
Rituximab MOA
Fab binds to the CD20 on B-lymphocytes & Fc, increases immune system for their lysis
Rituximab thera uses
NHL, CLL
Rituximab side effects
Arrhythmias, Hypersensitivity rxns (hypotension, bronchospasm, angioedema), Hepatitis B infections, Tumor lysis syndrome
Imatinib MOA
Inhibit tyrosine kinase, e.g., associated w/ Bcr-Abl mutation, hence ↓ proliferation & $ apoptosis

CML (Ph+), ALL (Ph+), GIST
