Ch 4: Autologous Conditioned Sera Flashcards
1
Q
What is autologous conditioned sera?
A
Incubated serum samples containing increased concentrations of IL-1ra.
Used to combat the IL-B2 pathway in the pathogenesis of OA
2
Q
How it autologous conditioned Sera made?
A
- 10-50ml whole blood collected with a large needle and excellend venipuncture technique
- slowly injected into a sterile, single-use chamber containing borosilicate beads
- Allowed to coagualte and incubated at 37degreesC for 6-24hr
- Leukocytes adhere to beads, induced cell signalling events leading to an increased expression and release of IL-1ra
- Centrifuges to isolate the serum
- Given as an intra-articular injection

Q
Literature…
A
- Management of human OA in 167 patients - no significant difference compared to placebo
- Management of OA of the knee in humans in comparison to hyaluronic acid. Significantly greater functional improvement up to 2yr post-Tx
- Horses treated with weekly series of 4 injection of middle carpal joint OA had significant clinical improvement and decreased synovial membrane hyperplasia and increased synovial fluid concentration of IL-1ra
