Cell Biological Techniques Flashcards
What are cell lines?
Typically derived from tumours
Abnormal when compared to primary cells
Easily transfected with nucleic acids
What are iPS cells?
Induced pluripotent stem cells
Reprogrammed somatic cells
Same properties of embryonic stem cells
Where is the gene of interest inserted?
Downstream of the promoter
How do cell biologists knock down gene expression?
siRNAs
What is cryo-electron tomography?
Enables visualisation of the molecular organisation of cellular components at near-atomic resolution
What is Flow cytometry?
Analyse the expression of proteins on the cell surface
Lazer excites fluorescent dyes at specific wavelengths.
Light emitted by the cells is measured by a series of detectors
What are FACs?
Fluorescent activated cell sorting, can sort populations. isolating single cells and nuclei for single cell genomics
What data can cDNA microarrays provide?
The relative levels of mRNA in a sample
What is SILAC?
stable isotope labelling with amino acids in cell culture. Differential labelling of cells with amino acids, typically lysine and arginine.
