What are the characteristics of BUA?
1) Final product of purine catabolism
2) Filtered by glomerulus
3) Reabsorbed and reused by PCT
4) Insoluble in plasma
What is the distribution (in percentage) of BUA (in terms of excretion)?
Urinary: 70%
GI tract excretion: 30%
What is gout?
It is the condition whereas there is high blood lvls of uric acid; deposition in joints
What is the other term for gout?
Gouty Arthritis
What are the components present if an individual has gout?
1) Inflamed joint
2) Swelling
3) Uric acid crystals
4) Deposit of uric acid (tophi?)
What are the purposes of BUA?
1) Confirm dx and treatment of gout
2) Prevent uric acid nephropathy during chemotherapeutic therapy
3) Assess inherited disorders for purine metabolism
4) Detect kidney dysfunction
5) Assist in dx of renal calculi
What are the sx considerations for BUA?
1) Heparinized plasma, serum, or urine
2) Non-fasting sx
3) EDTA or fluoride additives should not be used for uricase method
4) Urine collection must be alkaline pH 8.0
What are the sxs that can be used for BUA testing?
1) Heparinized plasma
2) Serum
3) Urine
Why should the urine sx collected be alkaline (having a pH of 8.0)?
To avoid crystals formation
What is the principle of rxn of BUA testing?
Uric acid + O2 + 2 H2O (via the action of uricase) -> Allantoin + CO2 + H2O2
2 H2O2 + 4-Aminoantipyrine + DHBS (via the action of peroxidase) -> chromogen + 4 H2O
What should be done after mixing the working rgnt w/ the sx?
Mix well and incubate for 10 mins
If spectrophotometric method is applied for BUA testing, what is the absorbance used for reading the sx?
520 nm
What is used as the blank for BUA testing and its corresponding volume?
Distilled H2O: 25 uL
What is used as the unknown for BUA testing and its corresponding volume?
Serum: 25 uL
