Blood and faecal samples Flashcards
Laboratory diagnosis of African trypanosomes?
chancre fluid aspirate
lymph node aspirate
blood sample
marrow
Concentration methods for African Trypanosomes?
Thick blood film
Woo concentration
Quantitative buffy coat-examined with fluorescence and microscope
Filariasis MF detection lab diagnosis methods?
Blood smears
thick blood films
Knott’s concentration of a sample
What is Knott’s concentration method?
Formalin added to blood, mixed and centrifuged, discard supernatant, add methylene blue, examine on slide
What is faecal antigen detection?
Monoclonal enzyme immunoassays for detection of specific antigen in faecal specimens- can use an ELISA
What are the two methods used for detecting parasites in faeces?
- direct wet mount microscopic examination
2. use of a concentration method followed by microscopic examination
What are the pros and cons of direct examination?
+
simple, quick, preserves trophozoites
-
often misses low numbers of eggs and cysts
What are the pros and cons of concentration methods?
+
increases sensitivity, removes fatty debris, fixes in formalin so preserves sample
-
time consuming, additional equipment, costly
What concentration techniques are there?
- flotation technique
2. sedimentation technique
Why might you get a false negative result from concentration methods?
- Intermittent shedding of parasites so might not be in the sample
- examining only one faecal sample
- naturally low number of eggs shed in faeces
- preparational errors
What is a McMasters chamber used for?
faecal egg counting- debris sink to bottom, eggs float to surface
slide is examined on a microscope and number of eggs in each chamber is counted
What culture methods can you use for Strongyloides?
- faecal charcoal cultures
- agar cultures(larvae moves through agar dragging bacterium with them)
What is the kato-kaz technique?
used to examine stool
What is the principle advantage of faecal concentration?
Increased sensitivity
Why is the concentration of faeces recommended for diagnosis of S.mansoni?
Low numbers of ova are produced (200 a day)
