What do cholinomimetics mimic?
Acetylcholine, primary NT of parasympathetic nervous system (Mime drink acetyl-cola)
What are the two type of receptors cholinomimetics use?
Nicotinic and muscarinic
Where are nicotinic receptors found?
Autonomic ganglia (transformers), skeletal neuromuscular junctions (plug box), and adrenal gland (beanie)
What type of receptor are nicotinic receptors?
Transmembrane, polypeptide acting like ion channels (TV channel in stores)
What type of muscarinic receptors are there? What are each coupled with?
M1, M2, M3. M1 with Gq, M2 with GI, and M3 with Gq. (Motorcycle parking spots, QIQ (quick) mart).
What proteins do each muscarinic receptor activate?
M1 and M3 (Gq) stimulates IP3-DAG cascade which lead to increased calcium (three puppies in motorcycle). M2 decreases cAMP (cAMP tent on back of motorcycle).
Where do you find each type of muscarinic receptor?
M1 found in CNS, brain, and enteric nervous system (brain helmet), M2 found in heart, specifically atria at the SA and AV notes (ripped heart shirt), M3 in smooth muscle and glands (smooth muscle bike and gland donut).
What physiological effect happen when M2 receptor stimulated?
Decrease HR at SA node, reduce contractility of atria, and reduce conviction velocity through AV node.
Do M3 muscarinic agonists vasodilate or vasoconstrict?
Vasodilate because they release NO which vasodilate (NO pipe on motorcycle).
What happen during arethrosclerosis?
There is vascular epithelial damage which results in increased cGMP and vasodilation.
Do muscarinics typically decrease or increase blood pressure in an average patient?
Decrease because they can cross to work on other M receptors in high doses.
What type of receptor does bethanechol use?
Muscarinic receptors (Beth drinking cola and cement pouring from colon sprout).
What does bethanechol treat?
Non-obstructive GI disorders (post-op ileus, neurogenic ileus, congenital megacolon). (DO NOT OBSTRUCT sign). Treats urinary retention as well (bladder hose).
What type of receptor does pilocarpine use?
Muscarinic (pile of carp).
What does pilocarpine treat?
Dry mouth (Sjogrens, radiation) by increasing salivation. Also treats acute angel glaucoma by causing meiosis (guys wearing constricted hoodie) and contracting the ciliates muscle to increase aqueous humor outflow (smooth muscle crane with net zonules).
