Astra Zeneca Flashcards
What is developability?
Assessing whether physicochemical properties and dose of API hinder or aid the development
What are the three mechanisms of powder mixing?
Convective, reducing scale of segregation
Diffusive, random motion required for microscopic mixing
Shear mixing, for cohesive particles
What are the three mechanisms of segregation?
Elutriation, smaller particles end up on top
Sifting, big particles end up on top
Trajectory
What three properties must be controlled during milling/granulating?
Tensile strength- maximised
Porosity- optimised
Granule PSD- minimised
What are the two types of powder flow regimes in wet granulation?
Bumping- impeller moving around, powder moving up and down, slow consolidation speed
Roping- good for dry mixing, high consolidation speed in wet granulation
What is hot melt extrusion?
API dispersed in polymer, barrel heated to melt polymer, extrudate cut into small particles for subsequent processing
What is the role of compaction pressure?
More compression reduces porosity of tablets, increasing disintegration and tensile strength
What is dwell time?
Time at which tablet punch exerts maximum pressure
What is pre-capping?
Splits/cracks on the sides of tablets due to excessive compression
What is capping?
Complete or partial segregation of tablet face
What is picking?
Removal of material from tablet face
What is a pan coater?
Spray guns used to spray coating solution onto tablet, hot air moves through to dry tablets
What is a fluid bed coater?
Typically used for pellet coating, spray concurrent to pellet flow and pellets fall to the bottom of the bed for re-entry into the tube
What is the purpose of wet granulation?
Produce larger agglomerates from fine powders
Improving product appearance and compressibility
Controlling solubility and porosity
What occurs if the bowl fill level is below the optimal?
Chopper redundant
Poor mixing
Slower consolidation rates
