Aseptic Technique Sheet For Lab Prac Flashcards
Describe molds?
Fluffy filamentous growth. Many colors possible
What is the difference in a mold filaments compared to streptomyces?
Folaments are of larger diameter than those of the prokaryotic Streptomyces filaments
What precaution should be taken with mold when plating them?
Plates with molds should not be opened; mold spores spread easily.
Describe Micrococcus?
Pigmented (yellow, red, orange, pink) glistening smooth colonies
They are small round cells
What Gram are micrococcus?
Gram + cell wall (stain purple) can be difficult to stain therefore they frequently look pink
Describe Bacillus?
Opaque cream or white colony
Relatively large, flat colonies which have a wavy edge
What gram is Bacillus?
Gram + cell wall structure (stain purple) but older cultures are gram-variable therefore might stain pink
What is one distinct feature about Bacillus and how they live/survive?
The are endospore forming
Describe Streptomyces?
Characteristic of an earthy smell or odor
Newer colonies are hard and white
Older colonies are black/brown in color, usually small
What is the shape of Streptomyces?
When viewed from the slide they look like a small volcano white base has been imbedded in the agar
Describe the composition of streptomyces?
Colonies are composed of mycelia filaments (not single cells) which stain gram positive.
Aerial filaments bear refractive (phrase bright), non-staining coindiospores
What produces ethyl alcohol and has a fermentation smell?
Yeasts
What gram stain are yeasts, an what is uniques about the cells?
Cells stain gram + and are larger than ordinary bacterial cells.
What does yeasts have that other bacteria don’t have?
Membrane bound organelles.
Mitochondria, distinct nucleus, and cytoplasmic streaming is eveident.
