what is aseptic technique
a way of growing micro-organisms cultures that greatly reduces a risk of contamination
why must we use aseptic technique when using micro-organisms?
to avoid the micro-organisms from being contaminated by air, workstations and apparatus. This allows us to only grow the micro-organisms that we want
what preparation must we do before we start and why?
-sterilise petri dishes
-sterilise work spaces
why do we work as close to the flame as possible
to avoid contamination (you guessed it)
and to enable a convection current
what do we use to inoculate out agar plates and liquid broth?
glass pipette or inoculating loop
why do we use ethanol during the technique
to sterilise the equipment
why do we need to take care of ethanol with the bunsen burners
ethanol is flammable
why do we need to shake our sample?
so that the sample has an equal proportion of bacteria in all places
why do we allow the ethanol to burn off/evaporate
because it could kill the bacteria that is needed
