Apoptosis Flashcards

1
Q

What happens in apoptosis?

A

The DNA fragments, the membrane blebs and the cell produces apoptotic bodies that produce “eat me signals” and the parts of the cell are then digested by phagocytes

Q

How can apoptosis be detected?

A

By TUNEL staining as chromatin become fragmented -the broken bits of DNA can be stained

Q

Mutations in TGF-B often cause cancer; why is this?

A

They cant undergo apoptosis and also they dont recognise the inhibition that TGFB tries to do

Q

What are the steps involved in producing a mouse knock out gene?

A
  1. Mouse totipotent stem cells
  2. Cloned copy of target gene
  3. Engineer knockout construct
  4. Transform ES cells with construct
  5. ES cells into ouse blastocyst
  6. Blastocyst into uterus
  7. Grow chimera mouse
  8. Breed knockout form of chimera
