Anxiolytics (PD - Week 3) Flashcards
What are anxiolytics used for
feeling of unease which can range from mild to severe
What are the effects of alcohol on GABA and Glutamate
increases GABA
decreases Glutamate
What regulates the stress responce
The HPA axis
Name 5 types of anxiety disorders
- General anxiety disorder
- specific phobias
- social phobias
- panic disorder
- OCD
- PTSD
Name 3 classes of drugs to treat anxiety disorders
- Benzodiazepines
- 5-HT1a receptor antagonists
- B-noradrenergic receptor antagonists
GABAa receptor allows the influx of which ion
Cl-
Where does GABA bind on the GABAa receptor
alpha and beta subunit
Where do Benzodiazepines bind on the GABAa receptor
alpha and gamma subunit
Explain the effect on GABA when benzodiazepines bind
enhances GABA inhibitory activity
Explain the relationship between tolerance and benzodiazepines
tolerance results in desensitisation of GABA receptors and an increased sensitisation of the excitatory neurotransmitter system NMDA glutamate receptors
Explain the relationship between benzodiazepines and withdrawal
decrease in GABA and increase in glutamate leads to heightened anxiety
What GPCR is 5-HT1a coupled to
Gi
Explain the mechanism of action of Buspirone
- partial 5-HT1a receptor agonist - inhibits serotonin release
- buspirone can cause desensitization of 5-HT1A receptor which leads to downregulation of this receptor and therefore increase serotonin
Explain the mechanism of action of SSRIs
- increase serotonin
- block the SERT transporter so more 5-HT is available
- used for long term potentiation
