Widely used
Subject to abuse
Many SA’s
Some are long acting- turned into other forms before being metabolized and excreted.
Some are short acting, and get eliminated quickly
Alprazolam
Triazolobenzodiazepine- intermediate acting for GAD, chemotherapy nausea
Diazepam
Sedative for Anxiety, seizures, AWS/BWS, muscle spasms, sleeping problems, RLS
Midazolam
used in ICU/CCU- anti seizure preanesthetic.
MoA of BZD
Bind between a&y and increase open frequency
MoA of BARB
Bind between a&b and increase open time
Carbamates
Meprobamate- popular after Barbs, replaced by bzds. Some countries have withdrawn it from the market. Still avaliable in the US, contolled though.
Agonist at GABA, narrow TI!
Buspirone
Agonist at 5HT1A r
GIRK Channels
Activated by GBy subunits from Gai/o receptors
Activation decreases RMP
Zolpidem
PAM at specific GABA receptors- not benzo or barb binding site
Melatonin
Agonist at melatonin receptor 1 and 2, GPCR coupled to Gai
