Reversibly inhibits 26S proteasome → activation of signaling cascades, cell-cycle arrest, & apoptosis
CCS: G2-M
Side fx: Reactivation of VZV (prophylactic acyclovir) and Hep B (possible fulminant hepatic failure)
Bortezomib
Selectively inhibits secretion of proinflammatory cytokines (ex: tumor necrosis factoralpha); enhances cell-
mediated immunity by stimulating proliferation of antiCD3 stimulated T cells; inhibits trophic signals to angiogenic factors in cells. Inhibits the growth of myeloma cells by inducing cell cycle arrest and cell death. Decreases VEGF.
Side fx: Myelofibrosis, ALL, Hodgkins, B-cell lymphoma, neutropenia, thrombocytopenia, peripheral neuropathy
Lenalinomide/thalidomide
Promotes T-cell proliferation
Cell cycle non-specific
*narrow therapeutic window, lots of side fx (general malaise, fever)
IL-2
Alkylating agent (new for this course), cross links DNA

SIDE FX: bone marrow supression
Melphalan
Inhibits purine synthesis/utilization and immunosuppressant (inhibits humoral response)

CSS: S phase

6-mercaptopurine
Synthetic glucocorticoid, anti-inflammatory, alters transcription, induction of cell death in immature lymphocytes
