Antineoplastic Flashcards
chemotherapeutic drugs
destroy or injure invading organisms or tissues: antimicrobial, antiparasitic, and antineoplastic drugs
antineoplastic drugs may distinguish cancer cells from normal cells based on:
- rate of growth and porliferation
- consumption of selected nutrients
- consumptoin of o2
what normal cells proliferate rapidly and are affected by antineoplastic drugs?
bone marrow - anemia, leukoplakia, infections hair follicles - alopecia buccal mucosa - stomatitis gonads - impotence embryonic tissue - teratogenicity
which types of cancer often have primary resistance to antineoplastic drugs?
malignant melanoma
brain cancer
rencal cell cancer
tumor lysis syndrome
side effects caused by debris from dead cancer cells
common with leukemia and lymphoma
complications of tumor lysis syndrome
hyperkalemia hyperphophatemia hypocalcemia hyperuricemia hyperuricosuira
what are the 3 methods of antineoplastic classification?
1) cell cycle: m=mitosis, s= DNA synthesis
2) mechanism of action: alkylating agents, antimetablites, microtubule inhibitors
3) chemical classification of antineoplasic: platinum analogues or steroid inhibitors
methotrexate MOA, Indications
antineoplastic
inhibits folic acid synthesis
I: lung cancer, breast CA, leukemia, Hodgkins and non-hodgkin lymphoma, cutaneous T cell lymphoma, osteosarcoma
SE: N/V/D, stomatitis, alopecia, myelosuppresion, peripheral neuropathy, heptaotoxicity
Doxorubicin MOA, etc
antineoplastic, antibiotic-like, anthracycline: inhibits DNA and protein synthesis
I: efficacious for broadest spectrum, IV rapidly, cardiotoxicity.
Cyclophosphamide MOA, etc
antineoplastic, alkylating agent, inhibits DNA replication, transcription of RNA, and nucleic acid function
I: acute lymphocytic leukemia, Hodgkins dz, non-Hodgkins lymphoma, breast, ovaria, lung cancers, mutiple myeloma, sarcomas SE: nausea, vomitting, diarrhea, abd pain
Paclitaxel MOA etc
antineoplastic and microtuble inhibitor
reversibly binds to microtubule preventing cell division
IV, Pacific Yew Tree, I: advanced ovarian ca, metastatic breast ca, SE: N/V/ appetite loss, joint pain, alopecia, fever, chlls, sore throat
Tamoxifin
antineoplastic hormone inhibitor/estrogen antagonist
binds estrogen receptor and blocks RNA synthesis
I: early breast ca, hot flashes, men sx, increased risk of uterine ca
