Antimetabolites Flashcards
1
Q
Antimetabolites general MOA
A
structurally resemble normal intermediates in the biosynthesis of purines/pyrimidines, but inhibit the enzymes involved in transforming that intermediate
2
Q
Folic Acid derivative agents
A
Methotrexate
Leucovorin
3
Q
Pyrimidine Analogs
5-FU
A
5-Fluorouracil
certain tumors used more uric acid. Inhibits thymidylate synthase (the adder of methyl group). Causes dose-limiting myelosuppression and cardiotoxicity.
4
Q
Methotrexate
A
blocks Dihydrofolate reductase. Affects S phase of cell cycle.
5
Q
Leucovorin
A
repletes tetrahydrofolate. Po. Rescue. Doesn’t require reduction by DHFR to synthesize pyrimidines. But..enhances cytotoxic effects of 5-FU.
