5-FU

A

5-Fluorouracil
certain tumors used more uric acid. Inhibits thymidylate synthase (the adder of methyl group). Causes dose-limiting myelosuppression and cardiotoxicity.

Methotrexate

A

blocks Dihydrofolate reductase. Affects S phase of cell cycle.

Leucovorin

A

repletes tetrahydrofolate. Po. Rescue. Doesn’t require reduction by DHFR to synthesize pyrimidines. But..enhances cytotoxic effects of 5-FU.
