Ketoconazole requires acidity for dissolution therefore H2-receptor antagonists, proton pump inhibitors and antacids decrease ketoconazole absorption

ketoconazole – hepatoxicity (rare), inhibition of adrenocortical steroids and testosterone (endocrine effects such as gynecomastia, decreased libido, impotence and menstrual irregularities,
