Antibodies Flashcards
What portion of current therapeutics is made up by antibodies?
1/3
What are the possible mechanisms of antibody action?
Ligand blockade, receptor blockade, receptor downregulation, depletion, signal induction.
How can antibodies use depletion?
Activate MAC complex, phagocytosis, lysis and pore formation.
Once hybridomas are produced, how can they be selected for?
Block the de novo synthesis of nucleotides but provide precursors for the salvage pathway. Myeloma cells cannot use the salvage pathway, but hydridomas can.
What mechanism do antibodies use to target TNF?
Ligand blockade.
What kind of antibody is Rituximab?
Chimeric anti-human CD20 antibody.
How does Rituximab function?
Bind to CD20 and activate complement pathways, NK cells and macrophages to kill B cells.
What kind of antibody is Adalimumab (humira)?
Fully human anti-TNF monoclonal antibody produced using guided selection.
How were plants altered to produced Zmapp (ebola antibody)
Lack specific plant N-glycans and contain mammalian enzymes.
