Antibodies Flashcards
Antibodies
Proteins that are part of specific immune system
Made in response to something detected as foreign
Specific: each Ab recognizes a distinctive antigen
Antigen binding sites: hand-like parts of Ab that attach to foreign bodies
Fc region: area of Ab at bottom on which fluorescent tags can be applied
Antigen
Target to which antibody binds
Triggers immune response
Epitope
Part of antigen to which antibody binds
Raising antibodies
Purify protein
Inject protein into host
Animal’s immune system will recognize protein as foreign
Animal produces antibodies
Blood drawn from animal will contain desired antibody
Monoclonal antibodies
Collection of identical antibodies
Polyclonal antibodies
Collection of monoclonal antibodies
Each group has different binding sites
Hybridoma cell
B cell injected with antigen A makes anti-A antibody but can’t divide forever
Tumor cell from cell culture divides indefinitely but doesn’t make any antibodies
Fuse B cell with tumor cell: hybrid cell makes anti-A antibody indefinitely
Uses of antibodies in cell studies
Determine how many of a certain cell type are in a population of cells Look for structural changes in a protein Measure amount of protein in a sample Drug delivery Locate protein Purify protein
