Antibiotic MOA Flashcards
Penicillins
Inhibits penicillin binding protein directly
Aminopenicillins
added side chains to increase the selectivity to Inhibit penicillin binding protein directly
Penicillinase Resistant Penicillins
Avoids binding with penicillinase/betalactamase so that it can still bind to the penicillin binding protein and inhibit it
Aminopenicillins/Betalactamse inhibitor
binds to betalactamase and penicillin binding protein and directly inhibits both
Antipseudomonal penicillins/betalactamase inhibitors
binds to betalacatmase and penicillin binding protein and directly inhibits both
Antipseudomonal penicillins
directly inhibits penicillin binding protein
1st gen cephs
Directly inhibits PBP
2nd gen cephs
Directly inhibits PBP
3rd gen cephs
Directly inhibits PBP
4th gen cephs
Directly inhibits PBP
5th gen cephs
Directly inhibits PBP
ceph/betalactamase inhibitors
Directly inhibits PBP
Carbapanems
Directly inhibits PBP
Monobactams
Directly inhibits PBP
Glycopeptides
indirectly inhibits PBP from crosslinking peptidoglycan by sitting on the side chains of bacteria
Tetracyclines
inhibits protein synthesis; 30s ribosomal subunit
Fluoroquinolones
inhibits both DNA gyrase and topoisomerase IV and prevents bacteria from replicating DNA
Macrolides
inhibits protein synthesis; 50S ribosomal subunit
Clindamycin
inhibits protein synthesis; 50S ribosomal subunit
Metronidazole
inhibits DNA synthesis by causing strand breakage
Bactrim
inhibits formulation of nucleic acids needed for DNA synthesis/replication
Aminoglycosides
Irreversibly binds with 30S ribosomal subunit causing the mRNA to be misread
Nitrofurantoin
metabolized within the bacterial cell to reactive intermediates that destroy ribosomal proteins
Oxazolidinones
inhibits protein synthesis; 50S ribosomal subunit
Daptomycin
binds with calcium and inserts itself into the phospholipid cytoplasmic membrane; other molecules aggregate and create holes in the membrane; this causes efflux of potassium and kills the bacteria
streptogrammin
Inhibits protein synthesis; 50s ribosomal subunit
Rifampin
Inhibits RNA polymerase; prevents RNA production
Fidaxomycin
Binds to RNA polymerase and doesn’t allow DNA to enter and transcription does not occur
Fosfomycin
inhibits enzyme MurA; causes death by inhibiting cell wall synthesis
