Q

how does aspirin work?

A

it irreversibly acetylates COX1/COX2 which therfore decreases synthesis of thromboxane

Q

name an ADP receptor inhibitor and its side effects

Q

explain how PDE inhibitors work

A

inhibition of PDE = increase cAMP in platelets = increase PKA = impaired platelet aggregation and formation of the platelet plug. NOTE INCREASE CAMP CAUSES ARTERIOLAR VASODILATION WHICH TREATS CLAUDICATION IN PATIENTS WITH PAD

Q

name 3 GP2b/3a inhibitors

A

abciximab, eptifibatide, tirofiban prevent platelet aggregation and plug formation. It can cause drug induced thrombocytopenia

7
Q

explain how thienopyridines work

A

they irreversibly bind to P2Y12 ADP receptor, which prevents aggregation of platelets. Eg. clopidogrel, ticagrelor, prasugrel. They are used for CAD, acute ischemic stroke, MI, decrease peripheral artery disease risk
