aging and cellular senescence Flashcards
Aging is not the same as
Development
Aging is characterized by 4 things
- External changes
- Physiologic and structural alterations in tissues
- Decreased ability to respond to stress
- Increased risk for age-related disease
Define aging
A collection of individual processes and ailments - outcome of complex interaction
- A coordinated and malleable process -
What 2 things make us think about aging as a coordinated and malleable process
genetics
caloric restriction
2 causes of aging
- Random molecular damage
2. Stress, poor diet, adverse environment
ROS react with
1 Fatty acids –> lipid peroxidases
- Proteins –> abnormal folding
- DNA –> mutations + breaks
Which organelle plays a role in aging
mitochondria
What is a cell’s limit to replication called
Hayflick limit
What is wener’s syndrome (not an exact model)
Accelerated aging due to a mutation in a DNA helicase
3 features of telomeres
Short repeat sequences
End of linear chromosomes
ensure replication of ends
Prevent fusion
DNA polymerase cannot completely replicate the
3’ end of DNA
5’ end of NEW strand is shortened
Telomerase is a
reverse transcriptase
adds DNA nucleotides
Telomere shortening is a defense against
development of cancer
Genes involves in slower aging and lifespan extension
IGF, and thyroid hormones, seen in mice, lower levels of GH and IGF-1
2 genes in mammals involved in lifespan extension
FoxO and sirtuins
