Pharmacology
What are the different subtypes of muscarinic receptors?
New Cocks Expel Semen Prematurely during Sex
M1 - Neural M2 - Cardiac M3 - Exocrine + Smooth muscle M4 - Periphery M5 - Striatal dopamine release
Give some locations of adrenoceptors and their organs.
See lectures and p778 table 61-1.
Draw the diagram for the control of HR with ANS?
See diagram
What blocks nicotinic receptors?
Trimetaphan and Hexamethonium
What blocks muscarinic receptors?
Atropine and Hyoscine
Atropine is used an anaesthetic premed to dry up secretions
What does the somatic nervous system control?
Skeletal muscle (including diaphragm and respiratory muscle)
What NS controls the vascular system?
SNS
a1 = vasconstriction
b2 = vasodilation
Describe the synthesis, release and metabolism of ACh?
See diagram
Choline acetyltransferase
Acetylcholinesterase
Describe the synthesis, release and metabolism of NA?
See diagram
Hydroxylation , decarboxylation, hydroxylation
COMT(target tissue) and MAO
What are the two types of ion channels?
Voltage sensitive
Receptor-linked
Give an example of a calcium channel blocker?
Nifidipine - used as LA –> generate fewer action potentials = reduced pain
What are the three ways drugs interact with enzymes?
Enzyme inhibitors
False substrates
Prodrugs
Give an example of an anticholinesterase?
Neostigmine
Give an example of a prodrug?
Chloral hydrate –> Trichloroethanol
insomnia
What is an example of protein binding drug?
Warfrin
Define affinity of a drug?
The avidity of a drug to its binding site
Define efficacy of a drug?
The ability of the drug to cause a conformation change in the receptor
Define full agonist
A drug that can produce the maximum response of a tissue
Describe cardiac glycosides?
Target the Na+/K+ pump system. They are cardiac stimulant drugs that slow Na+/K+ pump down leading to increased intracellular Ca2+ so an increased inotropic effect.
Give an example of a cardiac glycoside?
Digoxin
Give an example of a competitive antagonist?
Atropine and propanolol
Give an example of an irreversible antagonist?
Hexamethonium
What is propanolol used for?
Competitive beta blocker
