Urine Microscopic Examination Flashcards
Specimen should be examined while ___ or ______
fresh or adequately preserved
Required Refrigeration for Specimen Preparation are:
Precipitation of amorphous urates/phosphates, other non-pathologic crystals
may dissolved some crystals
Warming @37degC
Standard amount of urine
Usually 10 and 15mL
12mL is frequently used
Note to the report for specimen volume
If the urine volume is not adequate,
especially in pediatric patients
Centrifugation time for urine
5 minutes at 400 RCF *may vary at other hospitals or lab
Volume for Sediment preparation
0.5 mL and 1mL are frequently used
(uniform amount of urine sediment)
Volume of sediment examined
Conventional Glass Slide Method
* Recommended volume is 20uL (0.02 mL)
covered by a 22 x 22 mm glass cover slip
detects casts, ascertain the general composition of sediment
LPO
identification of urinary sediments
HPO
Casts have a tendency to locate near the edges of the coverslip (Low power scanning of the coverslip perimeter is recommended. Start with low brightness)
Conventional Glass Slide Method
Point of reference
Epithelial cells (big in size)
Average number per LPF
Casts
Average number per 10 HPF
RBCs and WBCs
Semi-quantitative (Rare, Few, Moderate, Many or 1+, 2+, 3+. 4+)
Epithelial cells, Crystals and other
sediments
